MSmtA4 is derived from the bacterial metallothione SmtA from Synechococcus PCC 7942 by mutating four sites, which effectively enhanced the adsorption capacity of cadmium, lead and other heavy metals. The SmtA sequence was derived from NCBI. The sequence of the family 2 CBM from endoglucanase A of C.fimi was obtained from NCBI. The sfGFP sequence is derived from the Snapgene database. Codon optimization was carried out to enhance the expression efficiency in the target strain. Linker between MSmtA4 and CBM: GSGGAGGS. Linker between CBM and sfGFP: GGGSPTGG.
